FIB-SEM volume imaging at room temp /cryo conditions

Method Specification

The entry of Salmonella in a distinct tight compartment revealed at high temporal and ultrastructural resolution.
J Fredlund et al. Cellul;ar microbiology 20 (4), e12816, 2018

Sequential cyclic removal of nanometric layers from the sample block face and the SEM image acquisition of each newly produced surface. In this manner, hundreds to several thousand of SEM images can be collected in automatic regime, and then processed to generate 3D dataset of the scanned volume. The typical volumes are from few microns to several tens of microns. Available for plastic embedded samples at room temperature, as well as for frozen samples at cryogenic temperature.
